        The iPure FFPE Nucleic Acid Kits utilize silica purification technology to extract high-purity total DNA or total RNA from paraffin-embedded tissue samples. After deparaffinization of the paraffin-embedded tissues with xylene or deparaffinization solution and digestion with proteinase K, the samples are transferred to a silica membrane by adding binding solution and ethanol to adjust the binding conditions. The silica membrane specifically binds DNA & RNA while proteins and impurities are not adsorbed and removed. After two rounds of protein removal and deionization, total RNA & DNA are eluted using Elution Buffer/DEPC water.

        1.Extraction of DNA and PCR amplification from human tonsil paraffin sections:

        DNA was extracted from paraffin sections of human tonsil samples obtained from the Pathology Department of the Third Affiliated Hospital of Sun Yat-sen University. Primers targeting the KRAS gene were used to amplify fragments of 107bp and 226bp, and the HBB gene fragment of 488bp.
